Resumo
The genus Eryhtroxylum belongs to the family Erythroxylaceae and is widely used in folk
medicine. Studies have shown that in this genus secondary metabolites of the classes of
alkaloids, flavonoids, steroids and terpenes were found. The present work was subdivided in
three stages, being: (i) study of the volatile organic compounds (VOCS) present in the leaves
of E. deciduum; (ii) study of the chemical profile of the leaves of four species of the genus
Eryhtroxylum (E. deciduum, E. suberosum, E. campestre and E. tortuosum) and cytotoxic
evaluation of the extracts against HeLA cells; (iii) chemical study and dereplication of the
ethanolic extract of E. deciduum leaves, and cytotoxic evaluation against 4T1 tumor cells. For the study of VOCS, the hydrodestilation and SPME techniques were used, presenting as main chemical composition, aliphatic hydrocarbons besides ketones, aldehydes, alcohols, esters and steroids. The chemical profile of the four species was performed using qualitative techniques such as CCD, HPLC-DAD, GC-EM and 1H NMR, evidencing the predominance of flavonoids, alkaloids and steroids. The dichloromethane and methanol extracts of E. campestre and E. suberosum; E. deciduum ethyl acetate and E. tortuosum methanol showed significant cytotoxic potential against HeLA cells (in vitro). The phytochemical study of the ethyl acetate fraction of E. deciduum led to the isolation of two glycosylated flavonoids: 4 ', 7-di-O-methylquercetin-3-O-β-rutinoside (I) and kaempferol-3-O-rutinoside (II), both confirmed by the fragmentation in analysis of sequential mass spectrometry and Nuclear Magnetic
Resonance experiments. The UHPLC-MS dereplication of the ethanolic extract of the leaves of E. deciduum led to the identification of four flavonoids, these being I and II, besides rutin (III) and catechin or epicatechin (IV), all already reported for the genus. This same extract was evaluated by in vitro assays against 4T1 type tumor cells, exhibiting inhibition similar to the cisplatin control. The results obtained are important for the knowledge of the studied species, since the literature presents very restricted data.
